By airplane: The nearest airport to Otranto is Brindisi (about 110 Km). Bari airport is farther (about 200 Km).

Duration of flight from some Italian airports:

 

Milan-Brindisi:  1,30 hour
Rome-Brindisi: 1 hour

By  train:The closest main railway station is in Lecce at about 50 Km from Otranto. To Lecce arrive the main trains.

Examples:

Milan-Lecce: 10 hours
Rome-Lecce: 6 hours

Then, to reach Otranto by train from Lecce, there is a local railway(it is not a direct line: you need to change 2 or 3 times), please check the timetable in this website:

Ferrovie Sud-Est (Southeast Railway)

By car: Take the A14 motorway ("Autostrada del Sole") and come off to the Bari Nord exit and follow the signs to Brindisi and Lecce along the SS16 ("Strada Statale SS 16 Adriatica).
